蛋白A(Recombinant Protein A)葡萄球菌细胞壁的组成成分,42kD,能特异性的结合免疫分子的Fc区,特别是IgG。有4个亲和结合位点,耐热和耐变性剂(4M尿素,6M盐酸胍)。具有不在抗原结合点而与免疫球蛋白结合的性质,能形成含有A蛋白、抗体、抗原的复合物,以湿菌液为原料制取。
重组蛋白A可偶联到固体分离介质(如agarose)上,用于单克隆抗体或多克隆抗体的纯化。重组蛋白A可与多种报告分子(荧光染料、酶标记、生物素、胶体金、放射性标记等)偶联并不影响其与抗体结合的活性。这些偶联后的蛋白A衍生物可用于Western-blot、ELISA、免疫组化等的抗体检测。
